Contents 1 Get the Lactate sensor to work and put out a reporter 1.1 Parts needed: 1.1.1 GBP - cloned into plasmid-still need to: change codons (site-directed mutagenesis-primer for each mutation) (turn it into lactate sensor) 1.1.2 TRZ (part just inside of membrane)-still need to get it from IGEM kit 1.1.3 ompR 2 Get the thermosensor working in a higher, more narrow range (38-40, 37-39) 2.1 Parts needed: 2.1.1 Toehold - we have the primers, we need to clone it into the plasmid 2.1.2 Plasmid - modify, so it has no SD and it has the proper MCS 2.2 Random Mutagenesis (sloppy polymerase) 3 Get the Cre-lox system working 3.1 Parts needed: 3.1.1 Cre-get a new template and doing PCR 3.1.2 Lox-PCR worked, but it still needs to be cloned into a plasmid 4 Modelling 4.1 Make a system of differential equations to describe the lactate sensor 4.2 Make a system of differential equations to describe the glutamate
