Get the Lactate sensor to work and put out a reporter

Parts needed:

GBP - cloned into plasmid-still need to: change codons (site-directed mutagenesis-primer for each mutation) (turn it into lactate sensor)

TRZ (part just inside of membrane)-still need to get it from IGEM kit

Get the thermosensor working in a higher, more narrow range (38-40, 37-39)

Parts needed:

Toehold - we have the primers, we need to clone it into the plasmid

Plasmid - modify, so it has no SD and it has the proper MCS

Random Mutagenesis (sloppy polymerase)

Get the Cre-lox system working

Parts needed:

Cre-get a new template and doing PCR

Lox-PCR worked, but it still needs to be cloned into a plasmid

Modelling

Make a system of differential equations to describe the lactate sensor

Make a system of differential equations to describe the glutamate
