BG-11 for Harvard/Golden in medium solid.
Reagents and Materials
- 100ml BG-11 Harvard/Golden liquid
- Antibiotics.
- 3% Agar.
Protocol
- Add 100ml of BG-11 Harvard/Golden liquid in a Erlenmeyer flask.
- Add 3g of agar in a flask wich contains BG-11.
- Set pH at 7’4 for Synechococcus elongatus to come from Harvard. In the other hand, set the pH at 8’9 for S. elongatus to come from Golden.
- Autoclave at 121ºC during 20 minutes while label and prepare the plates.
- Add the antibiotics. Cool BG-11 with the agar.
- Fill BG-11 in the plates and leave until solidify.
