Affinity chromatography

Procedure

Column preparation

Before use the column needs to be activated:

  1. drain the ethanol
  2. wash the column with dist. water
  3. load the column with 12 mL NiCl2 solution
  4. wash the column with PBS buffer

Now the column is ready to use.

Columns can be reused. In oder to store them:

  1. wash the coumn with EDTA buffer
  2. discard the flowthrough and wash two times with 20% ethanol
  3. close up the column with Parafilm and store at 4°C
