Affinity chromatography
Procedure
Column preparation
Before use the column needs to be activated:
- drain the ethanol
- wash the column with dist. water
- load the column with 12 mL NiCl2 solution
- wash the column with PBS buffer
Now the column is ready to use.
Columns can be reused. In oder to store them:
- wash the coumn with EDTA buffer
- discard the flowthrough and wash two times with 20% ethanol
- close up the column with Parafilm and store at 4°C
