- Jeremiah and Chris:
- Cultured BL21 cells (glycerol stock) containing the TBP+ insert in the L.A. expression plasmid (added B-complex)
October 22
- Jeremiah and Chris:
- Once an OD(600) was measured to be 1.0 the cells were expressed using an L.A. gradient
- An 8 hour and overnight time sample were taken
October 25
- Jeremiah and Chris:
- Cells were lysed using a freeze/thaw method to separate the inclusion bodies from the rest of the lysed cell
- Protein samples were prepared for SDS-PAGE
October 26
- Jeremiah and Chris:
- Run SDS-PAGE (70 V for 2.5 hrs)
- Stain gel with coomassie brilliant blue
