The insert to vector molar ration of 1:5 was chosen and caltulated with the formula:

Schmidt´sche thermocycler ligation program (STLP)

  1. T= 4.0°C 10 min
  2. T= 10°C 60 min
  3. T= 12°C 15 min
  4. T= 16°C 10 min
  5. T= 25°C 10 min
  6. T= 37°C 5 min
  7. T= 70°C 20 min
