Week 12 Resurrection

Pick up single colonies from plate of TA-9-2 and amplify them in LB liquid medium. Do colony PCR to identify the ligation procedure.

1. LIU Xiao continues his mutation D0 library but finds that colonies on plates strange. We suspect antibiotics on these plates are getting worse.

2. LIU Xiao made 6 half-finished( one arm long and the other short of D0 MS2 aptamer) and sent sequencing.

1. Dr. Song from Tianjin University visits us and raises morale in us. She advises us to show off our confidence and strives for modeling things.

2. We restart our lab meeting and plan for last month’s sprint.

Gap day……

Part Riboscaffold version 1’s sequencing results are so wrong %>_<%, so Yan makes part again.

LIU Xiao continues his mutation D0 experiments.

1. Chen and LIU Huachun make competent cells of BL21*DE3.

2. We restart cotransformation and coexpression of RNA scaffold+FA+FB.
