Cell Transformation
- Thaw Origami cells on ice
- Aliquot 50uL of competent cells into two pre-chilled 15mL culture tubes
- Add DNA (1-5ng in 1-5uL) to one of the culture tubes (other culture tube being a control)
- Swirl gently with pipette tip
- Chill tubes on ice for 30 minutes
- Heat pulse tubes at 42°C for 30-40 seconds
- Chill tubes again on ice for 2 minutes
- Add 500uL of LB media into each tube
- Culture cells in tubes in a 37° incubator while shaking
- Plate cultures on Amp/Kan plates and incubate at 37°C
