Cell Transformation

• Thaw Origami cells on ice
• Aliquot 50uL of competent cells into two pre-chilled 15mL culture tubes
• Add DNA (1-5ng in 1-5uL) to one of the culture tubes (other culture tube being a control)
• Swirl gently with pipette tip
• Chill tubes on ice for 30 minutes
• Heat pulse tubes at 42°C for 30-40 seconds
• Chill tubes again on ice for 2 minutes
• Add 500uL of LB media into each tube
• Culture cells in tubes in a 37° incubator while shaking
• Plate cultures on Amp/Kan plates and incubate at 37°C
