Week 14: 10th September - 16th September
this week we have worked extensively on a batch of different manipulations including:
- biobricking promotors in Psb1c3 which are:
- P Elastase
- Hsp70
- P Car A
- FLk1
- P CMV
- Golden gate assemebly of GFP-AID-Tir1 cassette in Psblc3 and Pcs2+: this included PCR amplification of seperate genes GFP-AID and Tir1 using then apllying the golden gate protocol over night, followed by trsnformtion, sequencing and characterization of those of PCS2+ in tadpoles
- Preparation of chemical Competent Cells
