Title

Transcriptional Terminator Efficiency Calculator (TTEC)

We used both theoretical model and experiment method. to study the rho-independent transcription terminators which regulate gene expression.

First, we developed a theoretical model to calculate the free energy of RNA folding and predict the secondary structure of terminators. An algorithm was proposed to predict terminator efficiency from the secondary structure.

We also constructed 100 terminators and did some wetlab experiments to validate our theoretical prediction. The basic method was measure the the terminator efficiency by measuring the GFP and RFP which are placed before and after terminators.

Besides,we developed a software and a web server for people to calculate terminator efficiency. We also built a database of terminator efficiency which we believe is the largest database of such kind.

Hope our work is useful for the comprehensive study on terminator efficiency.
