this week we have worked extensively on a batch of different manipulations including:

  • biobricking promotors in Psb1c3 which are:
    • P Elastase
    • Hsp70
    • P Car A
    • FLk1
    • P CMV
    this work included standard pcr amplification, purification , plasmid digestion, ligation of vector-insert, transformation, miniprepping, and sequencing.
  • Golden gate assemebly of GFP-AID-Tir1 cassette in Psblc3 and Pcs2+: this included PCR amplification of seperate genes GFP-AID and Tir1 using then apllying the golden gate protocol over night, followed by trsnformtion, sequencing and characterization of those of PCS2+ in tadpoles
