Inoculate O/N cultures for successful plate of Bs strain 1A833

• Inoculate one isolated colony (or as close as possible) into ~2 mL of LB+Spc media in a sterile culture tube

• 'For LB+Spc, 20 uL of Spcp stock (see above) was added to 2 mL of LB and mixed thoroughly before dispensing

• Incubate cultures overnight with shaking at 37^o C
