Make O/N cultures (3x) of pBR322 Ec XL1-Blue competent cells
- Performed as on Tues, July 17
- A plate was also inoculated from a robust colony to obtain a new fresh lawn of transformed XL1-Blue cells
Plate Bs strains for competence induction
- Plate Bs strains 168, PERM739, PY79, and 1A833 from glycerol stocks on appropriate media plates to obtain a patch of growth
- Incubate at 34o C overnight
