• June 22, 2012: Katie repeated the ADH amplification. A very thin band was cut from the gel.

Digestions were performed on ADH, mORANGE, GFP, and J63010.

• June 23, 2012: Katie ran the digestions in a gel. ADH showed no bands, probably too little template.

mORANGE, GFP, and J63010 were extracted from the gel.

• June 25, 2012: Akshitha repeated the amplification of ADH in six reactions using low temperatures and two different temperatures. Three of the six ADH reactions worked but one was too small to be cut from the gel. The other two bands were extracted. ADH was cut with E&P and with S&P.
