II. Made plain agar plates:

Protocol:
  1. Agar– 15 g
  2. LB broth – 25g (10 g Tryptone, 5 g Yeast Extract, 10 g NaCl)
  3. DI water – 1L
  4. Combine 1 through 3, and autoclave on “wet,” “liquid,” etc. cycle making sure to putthe solution in a bin about 1/3, 1/4th full with water
  5. Pour agar into sterile bottles for easier handling
  6. When the liquid is cool enough to handle, pour the solution into plates up to the arrow mark.
  7. Plates can be stored in a 4C refrigerator for about a month or longer

Note: agar can also be stored at 4C and reheated in a microwave or on a hot plate for use, but it may clump or become disrupted if ice crystals form.
