- PCR in large volume to have a lot a available biobricks
- Treatment with DPN1 to eliminate plasmids
- Gel purification to only keep our interest fragments
- Gibson Assembly method to put together the different biobricks for each construction
- Transformation with assembly products and staggering on a selective medium
- Colony PCR to have results before the transformation results
