8.20.12 Digested PSB1C3 + Yap + GFP + cyc, PSB1C3 + URE2 + GFP + cyc, and PSB1C3 + HAP + GFP + cyc with EcoRI & PstI for confirmation. Digested J63010 + Adh + mOrange with SpeI and PstI. Digested J63010 + pIRES + GFP + cyc with XbaI and PstI.

8.21.12 Ran gel of check digestions for PSB1C3 + Yap + GFP + cyc, PSB1C3 + URE2 + GFP + cyc, and PSB1C3 + HAP + GFP + cyc. All three constructs were confirmed. Ligated pIRES + GFP + cyc into J63010 + Adh + mOrange. Did 1:0, 1:10, and 1:30. Performed colony PCR on J63010 + Adh + mOrange + pIRES + GFP + cyc.
