The purpose of the 2012 Rose-Hulman iGEM team’s project is to to construct mating type tester strains that can be used to determine the mating type of an unknown yeast strain. We aim to reduce the needed time to accomplish the test from the time required for the current method, which is approximately three days. In order to accomplish this, we will first establish two different tester strains of yeast, one of each mating type. These strains will be modified with a positive feedback loop, activated by the mating process, that continuously produces fluorescent protein. Each tester strain’s fluorescent protein will be a different color so that both a positive and negative response can be obtained from the test. We hope that this project will help to facilitate future breakthroughs in yeast genetics. | |
