Monday
Making bacillus competent
Bacillus salts *10 made up:
- (NH4)2SO4: 20g
- KH2PO4 (anhydrous): 60g
- Anhydrous K2HPO4 189.7g (N.B. Different from the one below!)
- KH2PO4 60g
- Tri-sodium citrate 10g
- MgSO4•7H2O 2g
- Added distilled water to make 1000mL
- Autoclaved
Medium A base *10 made up:
- Yeast extract 10g
- Casamino acids 2g
- Added distilled water to make 900mL
- Autoclaved
bacillus strain 168 streaked out and grown on plate overnight.
