Experimenters: Jenn & Boris
Transformation of Multiple Kill Switches
Objective: To extract DNA from kit plates and transform into E. coli DH5
- K112806 from 2012 kit plate 4 well 6M
- K112808 from 2012 kit plate 3 well 6G
- K117000 from 2012 kit plate 2 well 14D
- K124014 from 2012 kit plate 3 well 20K
Procedure:
Extraction of DNA from kit plates
- 10 µL of Milli-Q H2O and extract parts from the kit plates Transformation into E. coli DH5
- Add 2 µL of DNA extract to 20 µL of E. coli DH5
- Incubate on ice for 30 min; 30 sec in water bath at 42˚C
- Immediately on ice for 5 min
- Add 400 µL of SOC media & resuspend
- Incubate for 1 hr at 37˚C in shaker
- Plate 200 µL of transformed E. coli on correct antibiotic plate
- Leave overnight in incubator at 37˚C
