Transformation of Multiple Kill Switches

Objective: To extract DNA from kit plates and transform into E. coli DH5

  • K112806 from 2012 kit plate 4 well 6M
  • K112808 from 2012 kit plate 3 well 6G
  • K117000 from 2012 kit plate 2 well 14D
  • K124014 from 2012 kit plate 3 well 20K

Procedure:

Extraction of DNA from kit plates

  • 10 µL of Milli-Q H2O and extract parts from the kit plates Transformation into E. coli DH5
  • Add 2 µL of DNA extract to 20 µL of E. coli DH5
  • Incubate on ice for 30 min; 30 sec in water bath at 42˚C
  • Immediately on ice for 5 min
  • Add 400 µL of SOC media & resuspend
  • Incubate for 1 hr at 37˚C in shaker
  • Plate 200 µL of transformed E. coli on correct antibiotic plate
  • Leave overnight in incubator at 37˚C
