The first step was to design and construct six overlapping oligos. These were added in a 1:1:1:1:1:1 ratio with each primer having a working concentration of 100pmol/uL. The following recipe was prepared:

|---|---|
| 20uL of each of the 6 primers | 120ul |
| 10X annealling buffer | 15uL |
| dH20 | 15uL |
| TOTAL | 150µl |
| 100X BSA | 0.5 µl |
| H2O | to 50 µl |
10X annealing buffer can be prepared using the following:
- 10mM EDTA
- 0.5M Tris HCl pH 8.0
- 0.1M MgCl2
- 0.5M NaCl
