pSB1C3-ccm clones 1 and 5, testing internal restriction sites. Lanes 1, 2 = EcoRI/SpeI digestion. Lanes 3, 4 = BamHI digestion. Lanes 5, 6 = ClaI digestion. Results appear as expected assuming 2 of the 3 ClaI sites are uncuttable due to overlapping dam methylation.
