The first step was to design and construct six overlapping oligos. These were added in a 1:1:1:1:1:1 ratio with each primer having a working concentration of 100pmol/uL. The following recipe was prepared:

Ingredient	Amount
20uL of each of the 6 primers	120ul
10X annealling buffer	15uL
dH20	15uL
TOTAL	150µl
100X BSA	0.5 µl
H2O	to 50 µl

10X annealing buffer can be prepared using the following:

- 10mM EDTA

- 0.5M Tris HCl pH 8.0

- 0.1M MgCl2

- 0.5M NaCl
