Today I did transformation of the following BioBricks: BBa_J23119 and BBa_I0462. First, I added 10 μL DW to their locations in the plate kit (plate No. 1). Second, I did the transformation of those BioBricks to component cells. The medium for the recovery was SOB (800 μL). Then, after shaking the cells for an hour at 37 degrees, I plated the cells on LB+Amp plates twice for each BioBrick, different volumes: 10μL and 100μL (4 plates in total). After that the plates were incubated overnight in the incubator 37 degrees.
