Ilya
- Restriction digest of Fus1 HF, Fus1 GC and Fus2 with EcoRI and PstI. We didn't digest Fus1 with XbaI and PstI because we used all of the DNA for the EcoRI and PstI digest. That's in order to make sure we have enough product for cloning into pSB1C3. Then we can get the insert from that plasmid for cloning downstream to BBa R0062 and BBa J23119.
- Made a glycerol stock of I0462 (freezer box position 1B) and J23119 (freezer box position 2B).
- Measured concentration of restriction products. Results: Fus1 HF - 29.6 ng/ul, Fus1 GC - 22.8, Fus2 - 41 ng/ul - relevant fragment concentration is 28.8 ng/ul.
- Set ligation for 1 hr at 30 degrees. Michal will put it in the 4 or -20 degrees freezer. I will do the transformation on Sunday.
