The LMU-Munich team is exuberantly happy about the great success at the World Championship Jamboree in Boston. Our project Beadzillus finished 4th and won the prize for the "Best Wiki" (with Slovenia) and "Best New Application Project".

Crust promoter measurements

The figure below shows the result of the evaluation of the three promoters for the spore crust proteins with a previous version of the luminescence reporter vector BBa_K823025. Each promoter (P_cgeA, P_cotV, P_cotYZ) has two independent clones and both were measured three times. To be able to know in which growth phase the cells are the optical density (OD₆₀₀) was measured throughout the whole experiment.

The result of the measurement of the three promoters P_cgeA, P_cotV and P_cotYZ. The left vertical axis displays the luminescence divided by the OD₆₀₀ and the left one shows the OD₆₀₀. On the horizontal axis the time is illustrated. The drawn through lines show the luminescence which correlates with the promoter activity and the dotted lines display the growth curve of each clone.

The native promoter of cotZ P_cotYZ and P_cotV show a signal in stationary phase whereas P_cotYZ is activated two hours earlier than P_cotV. It is displayed that these promoters are active for about 12 hours while P_cgeA does not show any acitvity.
