Chemical Transformation/Heat Shock Protocol from Bioline
- Remove and thaw chemically competent cells
- Aliquot 1-5µl DNA per 40-50µl of competent cells
- Incubate on ice for 30 minutes
- Heat shock cells for 45 seconds at 42°C
- Let cells stand on ice for 2 minutes
- Place cells in growth media and let it stand for 1 hour
- Plate cells on appropriate antibiotic and add IPTG when appropriate
