Chemical Transformation/Heat Shock Protocol from Bioline

  1. Remove and thaw chemically competent cells
  2. Aliquot 1-5µl DNA per 40-50µl of competent cells
  3. Incubate on ice for 30 minutes
  4. Heat shock cells for 45 seconds at 42°C
  5. Let cells stand on ice for 2 minutes
  6. Place cells in growth media and let it stand for 1 hour
  7. Plate cells on appropriate antibiotic and add IPTG when appropriate
