Nuclease assay

  1. E. coli genome was prepared using MP bio kit and the DNA was quantified using picogreen assay
  2. 2µg of DNA was put into each test condition.
  3. Appropriate buffer(s) were added for each enzyme/enzyme combination in the tubes.
  4. 10 units of enzyme was added into appropriate conditions
  5. At each timepoint aliquots were taken out of the reaction tubes and run on a 1% gel for 40 minutes and imaged.
