Make glycerol stocks (3x) of pBR322-transformed XL1-Blue O/N cultures

• Aliquot 600 μL of culture from successful tube (1 of 2) into 3 Eppendorf tubes
• Add 200 μL to each and vortex gently to mix
• Store at -80^o C

Plate Bs strains from glycerol stocks for electrocompetence protocol

• Plate Bs strains 168, PERM739, PY79, and 1A833 on their respective selection plates from glycerol stocks
• Incubate overnight at 34^o C
